Homogenize up to 24 aorta or other tough arterial tissue samples simultaneously-- 5 minutes or less!

 

The Bullet Blender™ enables you to homogenize up to 24 samples (aorta or other tissues) in separate microcentrifuge tubes at a time.  Load up to 300mg heart tissue with your application buffer and grinding beads in standard polypropylene microcentrifuge tubes.  Place the tubes into the Bullet Blender™ (pictured above) or Bullet Blender™ Blue.  The "bullets" in the "blender" vigorously strike all the sample tubes as the instrument runs for a few minutes.  The agitation of the grinding agents with the tissues homogenizes the samples.

The  Bullet Blender™  and Bullet Blender™ Blue enable you to disrupt or homogenize up to 24 aorta and other tissue/cell samples at a time.  Load the samples in standard polypropylene microcentrifuge tubes into the Bullet Blender ™.  Balls (the "bullets") repeatedly strike the sample tubes in a controlled manner, thousands of times per minutes, inducing vigorous motion of the tissue samples or other substances inside the tubes, providing efficient mixing. The samples are homogenized (or mixed) non-invasively.  The sample tubes are kept closed during agitation, as the samples are processed.  There are no probes to clean between samples.

Protocol for Aortic Tissue Homogenization in the Bullet Blender™

The protocol described in this document is for the use of the Bullet Blender™ for the homogenization of aorta. This protocol was developed using mouse aorta, and the time and speed settings may differ due to variations from species to species. This protocol was used for the extraction of proteins and does specify a buffer, however please choose whatever buffer is which is most appropriate for your application and modify this protocol as necessary.

Materials Required: epithelial tissue, Bullet Blender™, SB20 buffer, pipettor, microcentrifuge tubes, and 0.5mm stainless steel beads (part number SSB05).


Instructions

  1. Cut tissue into appropriately sized pieces for analysis (5mg-300mg) and place into a microcentrifuge tube. If possible, use long thin tissue pieces.

  2. Strip the outer fatty layer from the Aorta. OPTIONAL: Wash tissue 3x with ~1mL PBS. NOTE: This step removes external contaminants.

  3. Add stainless steel silicate beads (0.5mm) to the tube. Use a mass of beads equal to 3x your mass of tissue. One scoop of beads ≈ 250mg.

  4. Add 0.025mL to 0.6mL SB20 buffer (2 volumes of buffer for every volume of cells, minimum of 25μL), containing 20% SDS, 0.1M Tris, pH6.8, and 10mM EDTA).

  5. Close the microcentrifuge tubes.

  6. Place tubes into the Bullet Blender™.

  7. Set controls for SPEED 8 and TIME 3 minutes. Press Start.

  8. After the run, remove tubes from the instrument.

  9. Visually inspect samples. If homogenization is unsatisfactory, run for another two minutes at the SPEED 10.

  10. Proceed with your downstream application.

SAFETY NOTE!!!

When using a centrifuge to separate your homogenate from the debris and beads, make sure your tubes are balanced.
