Homogenize up to 24 blueberry samples simultaneously-- in less than 3 minutes!

 

The Bullet Blender™ enables you to homogenize up to 24 samples (blueberry or other fruit) in separate microcentrifuge tubes at a time.  Load up to 300mg blueberry (including skin and seeds) with your application buffer and grinding beads in standard polypropylene microcentrifuge tubes.  Place the tubes into the Bullet Blender™ (pictured above) or Bullet Blender™ Blue The "bullets" in the "blender" vigorously strike all the sample tubes as the instrument runs for a few minutes.  The agitation of the grinding agents with the tissues homogenizes the samples.

The Bullet Blender™ and Bullet Blender™ Blue enable you to disrupt or homogenize up to 24 berry and other tissue/cell samples at a time.  Load the samples in standard polypropylene microcentrifuge tubes into the Bullet Blender ™.  Balls (the "bullets") repeatedly strike the sample tubes in a controlled manner, thousands of times per minutes, inducing vigorous motion of the kidney samples or other substances inside the tubes, providing efficient mixing. The samples are homogenized (or mixed) non-invasively.  The sample tubes are kept closed during agitation, as the samples are processed.  There are no probes to clean between samples.

Blueberry:  Place about 100mg of fruit in each microcentrifuge tube. Load 100-150mg stainless steel bead blend (0.9-2.0mm dia) in each tube.  Add 100-200μL buffer (your choice, depending on your downstream application), then place the tubes into the Bullet Blender™.  Set the time to 3 minutes and the speed to 10.

Protocol for Blueberry Homogenization in the Bullet Blender

The protocol described in this document is for the use of the Bullet Blender for the homogenization of blueberry (flesh, seeds and skin form the genus Vaccinium L.). This protocol does not specify a particular buffer - you may choose which is most appropriate for your downstream application (nucleic acid isolation, protein extraction, etc.).

Materials Required:

blueberry, saline, aspirator, Bullet Blender™, homogenization buffer, pipettor, microcentrifuge tubes, and 0.9-2.0mm stainless steel bead blend (part number SSB14B).

Instructions:
  1. OPTIONAL: Wash blueberry 3x with ~1mL PBS. Aspirate. NOTE: This step removes external contaminants and debris.
  2. Section blueberry into quarters. Place quarter (100-200mg) into a microcentrifuge tube. Size may vary depending on species.
  3. Add a mass of stainless steel bead blend equal to 1.5X the mass of fruit. One scoop of beads ≈ 200mg.
  4. Add 0.2mL to 0.6mL buffer, i.e. 2 volumes of buffer for every mass of fruit.
  5. Close the microcentrifuge tubes.
  6. Place tubes into the Bullet Blender™.
  7. Set controls for SPEED 8 and TIME 3 minutes. Press Start.
  8. After the run, remove the tubes from the instrument.
  9. Visually inspect samples. If homogenization is unsatisfactory, run for another three minutes at the SPEED 10.
  10. Proceed with your downstream application.

SAFETY NOTE!!!

When using a centrifuge to separate your homogenate from the debris and beads, make sure your tubes are balanced.
