Homogenize up to 24 meconium samples simultaneously-- in less than 3 minutes!

 

The Bullet Blender™ enables you to homogenize up to 24 samples (meconium or just about any animal or plant tissue) in separate microcentrifuge tubes at a time.  Load up to 300mg meconium with your application buffer and grinding beads in standard polypropylene microcentrifuge tubes.  Place the tubes into the Bullet Blender™ (pictured above) or Bullet Blender™ Blue The "bullets" in the "blender" vigorously strike all the sample tubes as the instrument runs for a few minutes.  The agitation of the grinding agents with the tissues homogenizes the samples.

The  Bullet Blender™  and Bullet Blender™ Blue enable you to disrupt or homogenize up to 24 meconium or tissue/cell samples at a time.  Load the samples in standard polypropylene microcentrifuge tubes into the Bullet Blender ™.  Balls (the "bullets") repeatedly strike the sample tubes in a controlled manner, thousands of times per minutes, inducing vigorous motion of the tissue samples or other substances inside the tubes, providing efficient mixing. The samples are homogenized (or mixed) non-invasively.  The sample tubes are kept closed during agitation, as the samples are processed.  There are no probes to clean between samples.

Protocol for Meconium Homogenization in the Bullet Blender™

The protocol described in this document is for the use of the Bullet Blender™ for the homogenization of human meconium. This protocol was developed for the extraction and downstream analysis of various chemical analytes. You may need to alter this protocol to adapt it to your downstream application.

Materials Required:

meconium, Bullet Blender™, methanol, pipettor, 2.0 ml microcentrifuge tubes, bead spatula, and 0.5mm stainless steel beads.

Instructions:
  1. Accurately weigh 0.25 g of meconium and add to a 2.0 ml microcentrifuge tube. Try to keep the walls of the tube as clean as possible, and ensure the meconium sits as low in the tube as possible.
  2. Add 1.4 ml of methanol to every tube.
  3. Add one scoop (~0.07 g) of 0.5mm stainless steel beads to each tube.
  4. Securely close the microcentrifuge tubes.
  5. Place tubes into the Bullet Blender™.
  6. Set controls for SPEED 8 and TIME 5 minutes. Press Start.
  7. After the run, remove tubes from the instrument.
  8. Verify that all samples have a uniform appearance. A smooth semi-liquid should exist. If homogenization is unsatisfactory, run for another five minutes at SPEED 8.
  9. Proceed with your downstream application.
SAFETY NOTE!!!

When using a centrifuge to separate your homogenate from the debris and beads, make sure your tubes are balanced.

Bullet Blender™ 05* Homogenization Protocol for Meconium

The protocol described in this document is for the use of the Bullet Blender™ for the homogenization of human meconium. This protocol was developed for the extraction and downstream analysis of various chemical analytes. You may need to alter this protocol to adapt it to your downstream application.

Materials Required:

meconium, Bullet Blender™, methanol, pipettor, 5.0 ml centrifuge tubes, bead spatula, and 0.9-2.0 mm stainless steel bead blend.

Instructions:
  1. Accurately weigh 1.0 g of meconium and add to a 5.0 ml centrifuge tube. Try to keep the walls of the tube as clean as possible, and ensure the meconium sits as low in the tube as possible.
  2. Add 1.5 g (! 1/16 tsp.) of 0.9-2.0 mm stainless steel bead blend to every tube.
  3. Centrifuge all samples briefly to force the meconium and beads to the bottom of the tubes.
  4. Add 2.9 ml of methanol to every tube.
  5. Securely close the microcentrifuge tubes.
  6. Place tubes into the Bullet Blender™ 05.
  7. Set controls for SPEED 7 and TIME 5 minutes. Press Start.
  8. After the run, remove tubes from the instrument.
  9. Verify that all samples have a uniform appearance. A smooth semi-liquid should exist. If homogenization is unsatisfactory, run for another 3-5 minutes at SPEED 7.
  10. Proceed with your downstream application.
