I purchased a PrecisionPak for tough tissue/plant matter/small organism. Can I use the contents and protocol with soft tissue?
Yes. You can use the tough tissue kit for soft tissue homogenization. However, we recommend using lower speed and time settings to avoid over-homogenization, which will affect the integrity of analytes. Call us if you need assistance to determine the gentler Bullet Blender settings.
I purchased a PrecisionPak for soft tissue/plant matter/small organism. Can I use the contents and protocol with tough tissues?
No. The bead composition in the soft tissue kit is not ideal for achieving optimal homogenization of the tough tissues. Sub-optimal homogenization can result in poor yield of biomolecules.
MAGneat Extraction Kits Troubleshooting
If the process ends with poorly homogenized material…
Repeat the homogenization process.
If your results contain little or no DNA yield…
Keep the sample in the magnetic separation device when removing supernatant during binding or washing steps. While removing the supernatant, make sure to angle the pipette tip away from the pellet of beads, to avoid disturbing them. Ensure that the Binding Bugger, Wash Buffer 1, and Wash Buffer 2 have had Isopropanol and Ethanol added correctly.
If you find remains of MAGneat beads in elution…
Leave on the magnetic stand until elution buffer is clear. Centrifuge the eluate for 2-5 minutes at maximum speed. Remove supernatant free of beads.
If you have a poor 260/230 Ratio…
Centrifuge eluate for 2-5 minutes at maximum speed. Remove supernatant and dispense into new tube ready to measure.
If MAGneat beads are being lost throughout the extraction…
Increase magnetizing time throughout the protocol. Make sure while removing the supernatant to angle the pipette tip away from the pellet of beads to avoid disturbing it.
If you have issues with downstream applications due to ethanol carryover…
Increase air drying time to ensure MAGneat beads have dried before adding elution.
